The Muscular Dystrophy Campaign has organised the event at Stanley Palace, a 16th century building, renowned for paranormal activity.
The charity, which raises money for people suffering with a number of medical conditions associated with progressive weakening and wasting of muscles, is looking for people to help raise funds by being sponsored.
Stanley Palace is famous for its hauntings, particularly by the spirit of Elizabeth Warburton, known as The Lady.
She died in 1627 and has been witnessed walking around the building on various occasions.
The event, on Saturday, September 27, starts at 9pm and finishes at 5am.
It includes a tour of the building and the group will be joined by paranormal investigators and mediums.
Experiments will also be conducted using ghost hunting and psychic equipment.
A reservation fee of £25 is needed to take part, and the minimum sponsorship is £175.
